Metabolic fingerprinting: Using proteomics to identify proteins in gymnosperm pollination drops

Apr. 10, 2013 ? Proteins are vital parts of living organisms, performing a variety of essential functions such as DNA replication, catabolizing reactions, and responding to stimuli. The complete set of proteins expressed in an organism at a given time, under defined conditions, is known as the proteome. While the genome of an organism remains relatively stable, the proteome is remarkably dynamic, varying from cell to cell and even within a single cell and changing rapidly in response to developmental and environmental cues.

Proteomics is a powerful technique for examining the structure and function of the proteome. For some organisms, proteomics can uncover the relationship between DNA, RNA, and the production of proteins -- enabling the comparison of the genome to the proteome. For those organisms that have not yet been sequenced, proteomics facilitates the discovery and identification of proteins. In a new study published in the April issue of Applications in Plant Sciences, graduate student Natalie Prior and her colleagues demonstrate the suitability of proteomics in determining the composition of gymnosperm pollination drops.

"The biggest limitation in what we are doing is that there is no published gymnosperm genome," says Prior. "Most of the work on gymnosperms has been anatomical, histological, or morphological. The biochemical perspective is really lacking."

Mediating signaling between the pollen and the ovule, one role of the pollination drop is to provide a germination medium, which can be species specific. Additionally, in some species, anti-microbial proteins have been identified, suggesting that pollination drops provide protection in addition to acting as a landing spot for pollen grains.

"The proteins we are finding are really starting points for other research," says Prior. "We can identify these proteins, but there is a lot more research that can be done once we know what proteins are there."

Identification of the proteins found in pollination drops provides a metabolic fingerprint and thereby informs understanding of seed plant evolution. Comparing the proteomes of different species allows for identification of proteins, elucidating pollen-ovule interactions in gymnosperms.

"We are using proteomics to examine the biological relevance of the proteins that the pollen grain is exposed to in the drop," comments Prior. "It's fascinating to know if any of those proteins are consistent among groups of gymnosperms and what we can learn from that."

Researchers engineer 'protein switch' to dissect role of cancer's key players

Apr. 10, 2013 ? Researchers at the University of North Carolina at Chapel Hill School of Medicine have "rationally rewired" some of the cell's smallest components to create proteins that can be switched on or off by command. These "protein switches" can be used to interrogate the inner workings of each cell, helping scientists uncover the molecular mechanisms of human health and disease.

In the first application of this approach, the UNC researchers showed how a protein called Src kinase influences the way cells extend and move, a previously unknown role that is consistent with the protein's ties to tumor progression and metastasis.

"This rationally designed control of protein conformations represents a breakthrough in computational protein design," said senior study author Nikolay Dokholyan, PhD, a professor of biochemistry and biophysics. "We now have a new tool for delineating the activities of various proteins in living cells in a way that was never before possible."

The research was published online ahead of print in the Proceedings of the National Academy of Sciences. In the study, Dokholyan created a "switch" that would make a protein wobbly and unable to do its job unless it was flipped "on" by a drug called rapamycin, which would stabilize the protein and let it perform its function.

The approach is a simpler and more reliable version of a protein engineering system pioneered three years ago by Dokholyan and Klaus Hahn, professor of pharmacology at UNC, called rapamycin regulated or RapR. In the old approach, the switching mechanism depended on two proteins and the drug. The first protein -- the one the researchers wanted to study -- was given the RapR modification and put in cells in tissue culture. The second protein was placed in the cells as well, but simply floated around until the addition of drug caused it to latch on to the modification in the first protein and turn it on. The problem with the approach was that some cells would have a lot of the first protein and less of the second, or vice versa.

"It became the Achilles heel of the technique, because there was variability in the results due to the different ratios between the proteins," said Hahn. "What Dokholyan was able to do, which was extremely challenging from a protein engineering standpoint, was to combine the two parts into one."

Dokholyan and his colleagues took the two proteins and broke them apart into their individual components, structures called alpha helices and beta sheets. They then rewired them together to make a whole new protein where the parts could interact with each other. When researchers compared this system, called uniRapR, with the previous approach, they found the new one gave cleaner, more reliable and more consistent results.

They then applied the technique to study Src kinase, a protein involved in the metastasis or spread of tumor cells. Scientists had postulated that Src kinase plays a role in cell motility, but previous methods have not allowed them to isolate its activity from other similar proteins.

Working both in cultured human cells and in the model organism zebrafish, the researchers showed that turning on Src causes the cell to extend its edges as part of cell movement. Now that they have dissected the role of one protein, the researchers plan to look at a variety of other kinases to understand their roles in the development, progression, and spread of cancer.

The research was funded in by the National Institutes of Health, the National Institute of Environmental Health Sciences, and the National Cancer Institute. US expected to increase aid to Syrian rebels

WASHINGTON (AP) ? The Obama administration's next step in aid to Syrian rebels is expected to be a broader package of nonlethal assistance, but expanding from food and medical supplies to body armor and night-vision goggles, as the U.S. grapples for ways to stem the bloodshed from Syria's civil war.

However, President Barack Obama has not given final approval on any new package and an announcement is not imminent, a senior administration official said. The U.S. continues to oppose directly arming the Syrian rebels, in part out of fear that the weapons could fall into the hands of rebels like those who have allied themselves with al-Qaida in Iraq.

Secretary of State John Kerry hinted at the new nonlethal aid package this week, saying the administration had been holding intense talks on how to boost assistance to the rebels fighting forces loyal to Syrian President Bashar Assad.

"Those efforts have been very much front and center in our discussions in the last week in Washington," Kerry said Tuesday, a day before meeting with Syrian opposition leaders in London. "I'm not sure what the schedule is, but I do believe that it's important for us to try to continue to put the pressure on President Assad and to try to change his calculation."

During Wednesday's meeting, the Syrian opposition leaders asked Kerry and other top Western diplomats specifically for military equipment, according to a senior State Department official who was present.

Kerry told them that the U.S. was looking at different options to help the rebels, but made no promises about any specific types of future aid, said the official, who wasn't authorized to speak publicly on the meeting and demanded anonymity.

Kerry also urged the opposition to organize itself better and said he'd attend a meeting April 20 in Istanbul bringing together the Syrian opposition's big donor nations from Europe and the Arab world.

The United Nations estimates more than 70,000 people have been killed during more than two years of fighting between rebels and government forces.

Britain and France have already been shipping armor, night-vision goggles and other military-style equipment to the rebels.

Earlier this year, the U.S. announced a $60 million nonlethal assistance package for Syria that includes meals and medical supplies for the armed opposition. The aid package marked the first direct American assistance to the opposition forces trying to overthrow Assad. It was greeted unenthusiastically by some rebel leaders, who said it did far too little.

While the Obama administration continues to resist arming the rebels, the U.S. has said it would not stand in the way of other nations that decide to take that step.

Senior officials from the White House, State Department and Pentagon held a high-level meeting Friday that focused on Syria.

Among those who attended Kerry's meeting in London Wednesday were the Syrian opposition's interim prime minister, Hassan Hitto; Vice Presidents Suheir Atassi and George Sabra; Secretary-General Najib Ghadbian and the opposition's envoys to the United States and Britain.

Kerry then met one-on-one with Russian Foreign Minister Sergey Lavrov for talks on the Syrian civil war. The U.S. and Russia have frequently been at odds over the conflict in Syria, with Moscow opposing action at the U.N. Security Council that would increase pressure on Assad.

The State Department official said Russia offered no indication that it was softening its position on Syria. Kerry reiterated the U.S. preference for a political solution that includes Assad leaving power, the official said, and agreed to continuing discussing the situation in Syria with Lavrov and other top diplomats during an evening meeting of the Group of Eight industrialized nations.

Chrysler recalls more than 247,000 cars, SUVs

DETROIT (AP) -- Chrysler Group LLC is recalling more than 247,000 SUVs and cars for several problems, including faulty gas tank hoses, broken drive shafts and air bag warning lights that illuminate when there's no problem.

The largest of the recalls, posted Tuesday on the U.S. National Highway Traffic Safety Administration website, covers about 139,000 Chrysler 300, Dodge Charger and Dodge Challenger sedans from the 2011 and 2012 model years. The automaker says that faulty wiring can cause the air bag warning lights to illuminate.

Cars made from April 11, 2011 to Dec. 14, 2011 are included. Chrysler says it will fix the vehicles for free and will start notifying owners about the problem this month.

The recall includes 119,000 cars in the U.S., with the rest in Canada, Mexico and other countries.

Chrysler also is recalling 78,500 two-wheel-drive Jeep Liberty and Dodge Nitro midsize SUVs because the drive shafts can break.

The SUVs are from the 2007 and 2008 model years. Chrysler says in documents filed with NHTSA that a heat shield can drop and rub against the drive shaft, causing it to wear and break. If the shaft breaks, the SUVs will lose power and the side air bags may inflate. Chrysler says it doesn't know of any crashes or injuries from the problem.

Dealers will install a bracket to hold the shield in place at no cost to customers.

The recall affects 61,400 vehicles in the U.S., 13,700 in Mexico, 1,000 in Canada and 2,300 elsewhere. Also included is a sister vehicle to the Liberty that is sold outside North America, the company said.

Chrysler says it has received less than 40 complaints about broken drive shafts in the vehicles.

Also, the automaker is recalling about 30,000 small SUVs because a faulty hose inside the gas tank can cause them to run out of fuel.

The recall affects Jeep Patriot and Jeep Compass four-wheel-drive vehicles from the 2012 model year. If they run out of gas, the engines can stall.

Chrysler says it's aware of one crash but no injuries or fires due to the problem.

The company says the SUVs have saddle-style gas tanks that go around the rear drive shaft. Fuel is pumped from one side of the tank to the other through a hose. But Chrysler says some of the hoses weren't formed right in manufacturing, and they can stop the flow of gas.

The problem stems from a worldwide shortage last year of a nylon resin called PA-12. The shortage sent automakers scrambling for substitutes, one of which was used in the Compass and Patriot fuel tank hoses.

Chrysler says the hoses were made with a new material, which required a higher manufacturing temperature than PA-12. Some malformed tubes were shipped to the tank maker before the problem was found by Chrysler engineers. The manufacturing process was changed and the tubes now work properly, Chrysler said.

Dealers will replace the hoses free of charge. Owners will be notified starting next month.

The recall affects about 20,500 SUVs in the U.S. and 2,300 in Canada. The rest are in Mexico and outside North America. They were made from Oct. 18, 2011 to May 7, 2012.

Spokesman Eric Mayne said it's rare that this many recalls are made public in one day. Chrysler constantly tests its vehicles to find problems and fix them before customers have any trouble, he said.

Bed bugs are notoriously tough to exterminate?many populations have already evolved resistance to common insecticides. But they do have a non-chemical enemy: kidney bean leaves.

The leaves' microscopic hairs pierce the bugs' feet, trapping them after just a few six-legged steps. It's a traditional anti-bed bug measure in the Balkans [See Michael F. Potter, The History of Bed Bug Management, in American Entomologist]. And tests have shown the leaves to be remarkably effective, but only when freshly picked?meaning it's not the most practical tactic for city dwellers.

So researchers tried a synthetic alternative. They created molds of the leaves, and cast 158 replicas using various plant-like polymers. The synthetic leaf surfaces looked identical to the real leaves, each sporting tens of thousands of sharp, hooked hairs. But they didn't impale or trap bed bugs in the lab?they just snagged them momentarily, delaying their march toward blood. Those results appear in the Journal of the Royal Society Interface. [Megan W. The researchers are still hunting for the bean leaves' secret. If they discover it, they say it could be a nifty way to treat infestations without pesticides. As long as the thought of all those bloodsuckers trapped and squirming under your bed won't bug you all night.

Christian leader in Egypt blasts Islamist president

CAIRO (AP) ? The leader of Egypt's Coptic Orthodox Church on Tuesday blasted the country's Islamist president over his handling of the recent deadly sectarian violence, including an attack on the main cathedral in Cairo.

The remarks by Pope Tawadros II underscore rising Muslim-Christian tensions in Egypt. They were Tawadros' first direct criticism of President Mohammed Morsi since he was enthroned in November as the spiritual leader of Egypt's Orthodox Christians. They are also likely to fuel the political turmoil roiling the country for the two years since the ouster of autocrat Hosni Mubarak.

Tawadros also warned that the state of Egypt was "collapsing" and described Sunday's attack on the St. Mark Cathedral in central Cairo, which serves as the Coptic papal seat, as "breaching all the red lines."

Tawadros said Morsi had promised him in a telephone conversation to do everything to protect the cathedral, "but in reality he did not."

Asked to explain Morsi's attitude, Tawadros, who spoke in a telephone interview to a political talk show aired on the private ONTV network, said it "comes under the category of negligence and poor assessment of events."

On Sunday, an angry mob of Muslims threw firebombs and rocks at the Coptic cathedral in Cairo, leaving two people dead. One of the two was identified as a Christian.

The attack followed a funeral service for four Christians killed in sectarian clashes in a town north of Cairo early the day before. A fifth person, a Muslim, was also killed. It was the deadliest sectarian violence since Morsi came to office nine months ago as the country's first freely elected president.

Tawadros also criticized the president over his decision on Monday to revive a state body mandated to promote equality between Egyptians regardless of their religious and ethnic background. Morsi's decision was in response to the sectarian violence.

"Enough already of formations, committees and groups and whatever else," Tawadros said.

"We want action not words and, let me say this, there are many names and committees but there is no action on the ground," he added.

Morsi has strongly condemned the recent violence and said that he considered any attack on the cathedral to be an attack on him personally. He also ordered an investigation into the violence.

"Should we wait for instructions to start an investigation when something happens?" Tawadros said in response to Morsi's order. "Egypt's laws must be adequate to deal with the situation. This is a society that is collapsing. Society is collapsing every day."

The office of Morsi's assistant for foreign relations issued a statement shortly after the pope's remarks, saying that the "Egyptian presidency would like to affirm its full rejection of violence in all its forms, and under any pretext, and affirms that all Egyptians are citizens who should enjoy all rights and are equal before the law."

"The presidency further stresses that it will not allow any attempts to divide the nation, incite sedition, or drive a wedge among Egyptians under any pretense and that it is doing all it can to realize the sovereignty of law and hold the assailants accountable," the statement added.

Christians make up about 10 percent of Egypt's estimated 90 million people. Copts have complained for decades of discrimination and issues such as the building of houses of worship or inter-religious love stories often ignite Muslim-Christian violence.

But attacks against Christians have increased since Mubarak's 2011 ouster, including more attacks on churches and at times forced evictions of entire Christian communities from small towns and villages.

With Islamists politically empowered since the overthrow of Mubarak's regime, Christians have become increasingly worried about their freedom of worship and belief.

During Sunday's funeral service at the St. Mark Cathedral, mourners chanted against Morsi, calling on him to step down. Witnesses say a street brawl broke out when Coptic activists tried to stop traffic to stage an anti-government march.

A mob, described by witnesses as residents of the area, pelted the Christians with rocks and firebombs and fired birdshot at them, forcing them back into the cathedral complex. The mob outside and the Christians barricaded inside then exchanged rocks and firebombs for hours into the night Sunday.

Many of the Christians denounced what they called a lack of protection for the service. When police did arrive in greater numbers, they fired tear gas and gas canisters landed inside church grounds, causing panic among women and children, while people outside the church cheered.

Some firebombs thrown from near the church landed at a nearby gas station, while witnesses said some in the church lobbed firebombs at the crowd outside.

Police said they have arrested four implicated in the violence, but didn't provide details. Tawadros was not in the cathedral at the time of the siege.

Intel teases nextgen Xeon E7, E5 and E3 range,

Truth be told, not much has been announced on the consumer end at IDF Beijing earlier today, but Intel did tease us with upcoming refreshes of its Xeon E7, E5 and E3 families for the enterprise space. In chronological order we have the Haswell-based E3 with TDP as low as 13W, and it's coming in mid-2013. This is followed by the "Ivy Bridge-EP" E5 in Q3 and then the "Ivy Bridge-EX" E7 series in Q4, the latter of which boasting three times the memory capacity of its predecessor, along with Intel's Run Sure reliability feature. More details in the press release after the break if you're into these flavors of chips.

Also mentioned at the keynote were the now-available Atom S12x9 family for storage systems, as well as a couple of upcoming 22nm 64-bit Atom SoCs codenamed "Avoton" and "Rangeley," both of which are sampling now and are expected to launch in the second half of this year. We've actually already heard of the microserver-friendly Avoton from Facebook's Open Compute Project, whereas Rangeley for network infrastructures was also detailed around the same time; so again, hit up the press release for more details.

(AP) ? A top South Korean official says he misspoke when he told lawmakers there is an "indication" that North Korea is preparing for a nuclear test. But that doesn't change what Seoul has been saying for months: that Pyongyang has already prepared a tunnel for a nuclear blast and can use it whenever it wants.

When a lawmaker asked earlier Monday whether there was an indication of increased personnel and vehicles at the North's nuclear test site, Unification Minister Ryoo Kihl-jae said "there is such an indication." He said he couldn't say more because it involved confidential intelligence.

The comments were recorded on video, but Ryoo later told lawmakers he couldn't remember making them and didn't mean to say them. He said he was "startled" by reports carrying his earlier comments.

ZoneAlarm Extreme Security 2013


Many security vendors offer a three-tier product line: standalone antivirus, security suite, and mega-suite. Bonus features in a mega-suite can include backup, tune-up, password management, encryption, and more. ZoneAlarm Extreme Security 2013 ($89.95 direct for three licenses) occupies the mega-suite slot in Check Point's product line, but it's woefully lacking compared to the competition and even compared to its own 2012 edition.

This product's main window is identical to that of the base-level ZoneAlarm suite. The only visible difference is that the Tune-up link is no longer disabled.

Limited Features
The product comparison chart on the ZoneAlarm website indicates that ZoneAlarm Extreme offers PC Tune-up and keylogger protection, features not found in ZoneAlarm Internet Security 2013. However, keylogger protection isn't entirely ready. My Check Point contact explained that the company is "transitioning to a new homegrown keylogger technology that has less conflicts with third party software," and that at present keylogger protection is handles by the behavioral detection component.

Last year's version included a license for ZoneAlarm DataLock, a whole-disk encryption product, but that product is "being phased out." A new File and Folder lock product replaces it. However, that product it's not yet integrated into ZoneAlarm Extreme.

"ForceField" virtualization technology was a big part of the first ZoneAlarm Extreme. This technology worked to protect the browser from drive-by downloads and prevent any Web-based malware from making damaging changes to the system. A glowing browser window border served as a reminder of its protection. And... ForceField is out. "It was a support and development nightmare," my Check Point contact said. "A more compatible replacement technology [is] in the works, but [we] are still in the awkward transition phase."

Limited Tune-Up
The Tune-Up feature isn't installed along with the rest of ZoneAlarm Extreme, but the first time you try to run it, you're directed to a Web page that lets you download and activate it. I smelled a rat at this very first step. Yes, it offered a button to let me install now, but another button proposed that I "Update Now" for $9.95. The extra $10 you pay for ZoneAlarm Extreme gets nothing but this feature; now they want another $10?

It turns out ZoneAlarm's PC Tune-Up is a stripped-down and re-branded version of a tool from Large Software. This version focuses specifically on the Registry, omitting features like management of startup programs, scheduling, and getting rid of useless files.

When you launch the Tune-Up component it goes through four stages of Registry cleanup. It backs up the Registry, scans for problems, fixes found problems, and finally defragments the Registry. Expert users can pick and choose which problems to fix; most users will just fix everything. On the off-chance that a "fix" breaks something, you can revert to the backup created in the first step.

I ran a Tune-Up on my physical test system, running Windows 7. I hit a snag at the very first step, as the utility reported numerous errors backing up the Registry. After I clicked the error messages away, though, it reported that the backup completed successfully. It found and fixed hundreds of Registry problems and proceeded to defragment the Registry, after which I had to reboot.

Once I rebooted, I ran the Tune-Up again... and discovered that it had done nothing the first time around. It still displayed error messages during backup, still found and allegedly fixed the same problems, and still found the Registry in need of defragmentation. After a third run-through, for a sanity check, I contacted tech support.

They suggested I launch the Tune-Up application by right-clicking and choosing "Run as Administrator." That didn't make a lot of sense, given that I was using an Administrator account, but I gave it a try. This time it worked. I wonder how many ordinary users will run Tune-Up over and over, not realizing that it isn't doing anything?

Stocks edge higher ahead of earnings season

Stocks?ended modestly higher Monday, shrugging off an early decline, as investors waited to see whether big U.S. companies would deliver on expectations of strong earnings in 2013.

Alcoa became the first major U.S. company to report earnings late Monday, and the results were mostly good. Aluminum maker's income was higher than analysts were expecting, but its revenue fell slightly short of expectations. Later this week the pace picks up with reports from Bed Bath & Beyond, Wells Fargo and JPMorgan Chase.

A big factor driving the Standard & Poor's 500 up 9.6 percent this year has been optimism that it will be a good year for company profits. While the expectations for the first quarter are relatively modest, many investors are expecting to see more of a pickup in earnings later in the year.

"We need to see some earnings growth here to justify the big gains we've seen in the first quarter," said Ryan Detrick, a senior technical analyst at Schaeffer's Investment Research.

Earnings for companies in the S&P 500 index are expected to rise by 0.7 percent from the first quarter of last year, but that growth is expected to accelerate sharply to 13 percent in the final three-month period of the year, according to data from S&P Capital IQ.

On Monday the Dow Jones industrial average rose 48.23 points, or 0.3 percent, to close at 14,613.48. The index started the day lower and fell as much as 67 points during morning trading. Alcoa's gain of 1.8 percent was one of the biggest in the Dow. It rose 15 cents to $8.39. The?stock?was off seven cents in after-hours trading.

The S&P 500 index closed up 9.79 points, or 0.6 percent, at 1,563.07.

The Dow and S&P have struggled for direction over the past three weeks. The S&P 500 has alternated between gains and losses every day as investors take advantage of any weakness to add to their holdings.

Telecommunications?stocks?fell 0.5 percent and health care?stocks?inched up just 0.2 percent, lagging the rest of the market. The two industry groups have performed well this year as investors sought out less risky?stocksthat pay good dividends. Health care companies are up almost 16 percent, making them the best performers in the S&P 500.

Lufkin Industries, an oilfield equipment maker, surged $24.03, or 38 percent, to $87.96 after General Electric Co. agreed to buy the company for $3 billion. GE wants to bolster its oil and gas operations. Its?stock?rose 19 cents, or 0.8 percent, to $23.12.

Johnson & Johnson logged the biggest percentage decline on the 30-member Dow Jones industrial average, dropping 93 cents to $81.11. Analysts at JPMorgan cut their rating on the?stock?to "neutral," saying it has risen too far, too fast. Johnson & Johnson is up 16 percent this year.

Stocks?fell Friday after the government reported a slowdown in hiring that was far worse than economists had expected. The report capped a bad week: The S&P 500 logged its biggest weekly decline of the year as signs emerged that U.S. growth is starting to cool.

In other trading, the Nasdaq composite index rose 18.39 points, or 0.6 percent, to 3,222.25.

The yield on the 10-year Treasury rose to 1.75 percent from 1.71 percent late Friday. It went as 1.69 percent Friday, its lowest level of the year. The benchmark rate has fallen from a recent high of 2.06 percent reached March 11 as demand for low-risk assets increases.

Fire official: 2 trapped North Carolina children likely dead

DENVER, N.C. (AP) ? Two young children trapped when dirt fell on them at a home construction site Sunday were not expected to be found alive and crews expected to work through the night to recover their bodies, a fire official said.

A father of one of the children called 911 at about 6 p.m. to report what happened, said Lincoln County Emergency Services public information officer Dion Burleson. Crews were on the scene in minutes, but couldn't get to the 7-year-old boy and 6-year-old girl, Burleson said.

Crews used shovels and climbing gear trying to get to the children at the Denver neighborhood. Emergency personnel from several places, including nearby Charlotte, were on the scene late Sunday.

Video from WSOC-TV shows backhoes scooping dirt from a deep hole surrounded by dirt. Equipment surrounded the hole. Burleson said it is hard to estimate how far down the children might be.

The man who called was the father of at least one of the children, Burleson said. Neighbors told WBTV that the children were cousins.

This is "devastating for both the family members and responders who are on the scene," Burleson said. "This is a tragic night in Denver."

Uganda News | Wildlife 'faces' promise to guard environment

Moses Waibi, a Senior Four student at Kira College Butiki in Jinja and Gadaffi Nawal, a Senior Five student of Kisaasi College in Kampala, will take two days off their books for a trip to Addis Ababa, Ethiopia.

This is after the duo was crowned Mr and Miss Wildlife 2013 during the Environment Education competition at Kitante Hill School at the weekend, instantly winning free fully-sponsored trips.

?Wildlife clubs enable students appreciate what they learn and it is a basic requirement for education,? Mr Fagil Mandy, the chairperson of the Uganda National Examinations Board, said shortly before crowing the winners.

Mr Mandy said schools should promote formation of clubs to boost co-curricular activities to enable students have a different understanding on various aspects of life.

Joining cause

After being crowned, Waibi said he would use his position to promote environment education in schools while Nawal said she would be an environment ambassador in schools across the country.

The two beat 98 students in a hotly-contested competition that covered general knowledge, thematic questions, and general presentation on the environment and wildlife.

Mr Dan Nuwamanya, an environment don at Makerere University, Ms Patricia Adoki, a Biology and science lecturer at Gulu University and Ms Claire Wandera, an education officer at Uganda Wildlife Educational Centre, were the judges.

The Vice Chancellor of the East African University and also board member of the Uganda Wildlife Clubs of Uganda, Prof. Eric Edroma, said they had come a long way in conserving the environment. He compared the sector to 1975 when poaching was at its peak to what the country can pride in today.

Mr David Musingo, the chairperson of the Wildlife Club of Uganda said plans were underway to organise regional competitions.

Early intervention is essential in the treatment of this disorder

Researchers of the Psychiatry and Mental Health research group at the Bellvitge Biomedical Research Institute (IDIBELL), in the Bellvitge University Hospital, have shown that patient age influences the onset of pathological gambling disorder and its clinical course. The study results were published in the Journal of Gambling Studies.

Personality traits

The study was conducted with more than 2,300 patients aged from 17 to 86 years. The coordinator of the study, Susana Jimnez-Murcia, explains that some personality traits associated with age are risk factors in different stages of life.

Younger patients (from 17 to 35 years) are more likely to be impulsive and seek new sensations that act as precipitating factors of gambling. In contrast, for older patients (from 55 to 86 years) these risk factors decrease but others increase, like harm avoidance (tendency to anxiety and excessive worry) and cooperation. "Older people do not gamble to seek awards or for the challenge of winning or for competitiveness as young people do, but they gamble to modulate negative emotional states", explains Jimnez-Murcia. "Older patients try to flee from loneliness, dissatisfaction or even physical discomfort."

"The only personality factor that does not vary with age is self-direction, the ability to take control of our lives, to be persistent with a goal" says the researcher. The research confirms that this factor "is low at all stages and that could be one of the keys to the treatment and prevention of the disorder."

Early intervention

The study associates young patients with higher severity in their gambling conduct. "Early intervention in these patients allows us to understand the causes of the problem in the early stages and we can give to patients tools to control it." One of the conclusions of the study is that pathological gambling is a temporary and episodic condition in patient's life. "We observe," says Jimenez, "that at certain times of the life, some social, environmental and personal factors can ease to control this behaviour but in other moments it is not so easy, and the psychopathology could be more serious. But the good news is that it is not a chronic disorder for life".

In patients aged from 55 to 86 years, the disorder is associated with health problems, while middle age is associated with economic problems. The study also detected a later gambling onset in women, from 35-40 years, while men begin to gamble younger.

Researcher Susana Jimnez Murcia explains that in recent years the profile of the patients has changed as a result of the rise of online gambling: "now we see younger, college-educated and higher socioeconomic level patients. We must continue working to promote responsible gaming and also to inform of the potential risk of gambling."

CAIRO (TheBlaze/AP) -- Al-Qaeda's leader has urged Muslims in "Arab Spring" nations to unite and institute an Islamic state, or caliphate, while warning France that its intervention in Mali will be bogged down.

"I warn France that it will meet in Mali, with God's permission, the same fate America met in Iraq and Afghanistan," Ayman Al-Zawahri said in the 103-minute audio message.

In the recording, al-Zawahri urged Muslims to liberate their lands from oppressive regimes and foreign troops, apply Islamic law, halt the plundering of Muslim wealth, support rebellious Muslims and oppressed people worldwide, and establish the Islamic Caliphate, or religious state.

The audio was produced by al-Qaeda's media arm, As-Sahab, and was posted on militant websites late Saturday.

"The enemy has begun to reel and collapse," he said, encouraging his followers.

The al-Qaeda leader also praised the rebels, or "holy warriors," in Syria, urging them to step up their fight against the regime of President Bashar Assad. But he also warned them against letting the country fall under the influence of the United States, the Arab League, the United Nations and Israel should they gain control of it.

"(They) want to steal your sacrifices and your jihad to give them to their supporters in Washington, Moscow and Tel Aviv."

Al-Zawahri also lashed out against Lebanese militant group Hezbollah and Iran for their support of Assad, saying that "the true faces of Iran and Hezbollah have been exposed, and their ugly reality has appeared in the field of holy war in Syria." He called The Syrian government a "criminal secular" regime.

Al-Zawahri, an Egyptian and former top deputy of Osama bin Laden, further criticized the ruling Muslim Brotherhood in Egypt for its weak response to the country's poverty, saying "the rich are getting richer and the poor are getting poorer. Have the Islamic movements provided better education, health or transportation?"

He also attacked the country's new constitution, drawn up by the Brotherhood and other Islamic movements, for not being religious enough.

But recent reports indicate Egyptian President Mohammed Morsi and al-Zawahiri may be working together on a plan to bring the terrorist back to his native country.? Al-Zawahiri traces his roots back to the Brotherhood, and his return would undoubtedly alter the goals and tactics of some of its members.

The al-Qaeda leader has not released a similar message since November.

Microsoft to Patch IE10 Pwn2Own Bugs Next Week

Oh yes, oh yes, it?s Patch Tuesday once again and Microsoft has nine security updates on tap for your downloading pleasure, two of them deemed critical. The download will also finally address the Pwn2Own bug, discovered last month at the Pwn2Own hack-a-thon.

Google and Mozilla patched the vulnerabilities disclosed in their Chrome and Firefox browsers within hours of the contest, but Microsoft has yet to issue a post-Pwn2Own fix for IE.

Gaza group: Hamas shaves heads for 'indecent' hair

GAZA CITY, Gaza Strip (AP) ? Police in Hamas-ruled Gaza have started grabbing young men with long or gel-styled spiky hair off the streets, bundling them into jeeps, mocking them and shaving their heads, two of those targeted and a rights group said Sunday.

It is the latest sign that the Islamic militants are imposing their strict practices on the population.

Hamas has been slowly forcing its fundamentalist interpretation of the religion on already conservative Gaza since it overran the territory in 2007, but the new crackdown on long hair and tight or low-waist pants ? in several cases accompanied by beatings ? appears to be one of the most aggressive phases of the campaign so far.

It began last week, and two of those targeted told The Associated Press said they were rounded up in separate sweeps in Gaza City that included more than two dozen young men.

House painter Ayman al-Sayed, 19, had shoulder-length hair before police grabbed him and shaved his head Thursday.

"The only thing I want to do is leave this country," said al-Sayed, who despite his ordeal defiantly wore stylish but outlawed narrow-leg tan khakis Sunday. "I am scared. They just take you from the street without reason. I don't know what they are going to do next."

Hamas officials played down the campaign ? a stance adopted in the past that allows the group to distance itself from a controversial crackdown while at the same time instilling fear in those it targeted.

Ziad al-Zaza, the deputy prime minister of Gaza, said the head-shaving "was a very limited, isolated behavior of the police and is not going to continue."

The Palestinian Center for Human Rights called on Hamas to investigate the "arbitrary detentions and violations of civil rights of civilians."

The hair crackdown came just days after the Hamas-run parliament in Gaza passed an education bill mandating separate classrooms for boys and girls from the age of nine.

Gender separation is already widely practiced in Gaza schools, as it is in the West Bank, where Hamas rival Mahmoud Abbas, the Western-backed Palestinian president, administers some areas.

Enshrining such separation in law marked another step forward in Hamas' campaign of imposing Islamic practice.

Since seizing Gaza from Abbas six years ago, Hamas has moved gradually in spreading its ultra-conservative version of Islam. It has issued rules restricting women or requiring them to cover up in the traditional Islamic dress of long robes and headscarves, but relented if met by protests.

Last month, the Hamas government barred girls and women from participating in a U.N.-sponsored marathon, prompting a U.N. aid agency to cancel the race. Hamas activists have also exerted social pressure to get all school girls to wear Islamic dress.

Al-Sayed, the house painter, and 17-year-old high school student Tareq Naqib said Sunday that they were targeted by police in separate incidents Thursday.

Al-Sayed said he had just finished his work in Gaza City and was waiting at an intersection for a shared taxi when a police jeep approached. Al-Sayed said he was thrown into the jeep with more than 10 others already squeezed into the back of the vehicle. He said policemen cursed them on the way to the police station.

There, the detainees were lined up, and a policeman began shaving their heads. He shaved two lines, from front to back and from one ear to the other, telling the young men they could finish the job at a neighborhood barber shop.

Those who resisted were beaten, al-Sayed said. He said he asked the policeman to finish the job of shaving so he wouldn't have to step outside with a partially shaved head.

A young man came into the police station, saying he was looking for his cousin, said al-Sayed. One of the officers grabbed the young man, who had his hair in gel-styled spikes, and shaved his head as well.

Naqib, the high school student, said he was seized outside his home and put in a police jeep along with four young men who had come to Gaza City from the southern town of Khan Younis.

On the way to the police station, police insulted them and warned them that Gaza is Islamic, said Naqib.

"They said, 'we want you to respect our tradition,'" Naqib said. "They made a cross on our heads and asked us to leave and finish the shaving at a barber shop."

Naqib's family is originally from Tunisia, and he said he wants to go back there after he finishes high school.

In another incident, a Gaza teen, who spoke on condition of anonymity for fear of retribution, said he saw police beat three young men in downtown Gaza City for wearing tight, low-rise pants. The witness said the policemen beat the three with clubs on the backs of their knees and told passers-by watching the scene to move along.

Ahmed Yousef, a Hamas figure identified with the more pragmatic wing of the movement, said the police behavior is "absolutely wrong" and must stop. Hamas is often divided over such campaigns, but the pragmatists have been unable to stop the more zealous members.

Hamas is also competing with the even more fundamentalist Salafis, a movement that has gained in strength and popularity in Gaza in recent years. Salafis have criticized Hamas for not implementing Islamic law in Gaza quickly enough.

For Gaza's young generation, such crackdowns have meant a shrinking space of self-expression. In some, it sparked defiance. Mohammed Hanouna, an 18-year-old high school senior, said he started styling his hair with gel after his friends Ayman and Tareq were targeted by police.

On Sunday, he walked with them in the streets in a show of support, adding that he is not afraid of arrest. "I have nothing to lose except my hair," he said.
